The France defender has tested positive for covid and has been forced into self-isolation.
Varane will have to stay away from the Madrid team for nine key days in which Real Madrid play in the Champions League against Liverpool and in the LaLiga against Barcelona.
Eder Militao is expected to be drafted in as a replacement for Varane tonight.
