Left back Nathaniel Brown was unable to take part in Germany's final training session before Thursday's last World Cup group match against Ecuador.
The Eintracht Frankfurt player completed an individual session in Winston-Salem due to minor groin complaints.
The 23-year-old is therefore not expected to feature in the game in East Rutherford, with Germany already assured of top spot in Group E after two wins.
Brown delivered outstanding performances as a starter against Curaçao (7-1) and Ivory Coast (2-1). His back-up against Ecuador is set to be RB Leipzig's David Raum.
All other 21 outfield players and four goalkeepers took part in the final session at the team base in North Carolina. The German team led by head coach Julian Nagelsmann were flying to New Jersey on Tuesday evening.
The match could be crucial to the South Americans but Germany may make other changes given they are safely through to the last 32.
